Abstract  The working principle and structure characteristics of fluorescent quantitative PCR analyer is introduced, and a calibration method based on optical simulation is put forward. Using variable intensity of PCR temperature calibrator and PCR fluorescence calibrator, and arranging the temperature sensor and LED light source, the fluorescence quantitative PCR analyzer temperature control performance and quantitative accuracy are calibrated, and the uncertainty components which may influence the results of the calibration are introduced.
